ImmunoSpot® test kits are carefully optimized to deliver maximum sensitivity for detecting and analyzing the secretory footprint of individual T and B cells, as well as to enable clear assessment of co-expression patterns.
Using a 4-color ImmunoSpot® kit, CD8+ T cell subpopulations can be simultaneously identified by measuring IFN-γ, TNF-α, GzB, and IL-2. (Learn more). See kit.
For CD4+ T cell subpopulations, Th1, Th2, Th17, and Th21 can be characterized based on their secretion of IFN-γ, IL-4, IL-17, and IL-21, respectively. However, multiplex analysis of these subsets must take into account their differential activation kinetics. (Learn more). CTL provides PBMC/antigen pairs that show Th1, Th2, Th17, and Th21 reactivity, available as positive controls and reference samples (please inquire).
Multiplexed detection of B cells secreting various immunoglobulin classes and/or IgG subclasses is also possible with up to 4-color kits. Additionally, we offer support in developing antigen-specific B cell assays by formulating the most appropriate coating conditions (please inquire).

All our kits are developed and standardized to eliminate variability and reduce subjectivity in performance. Our T cell kits include serum-free assay medium, removing the need for blocking steps and eliminating batch-to-batch variability common with FBS-containing media in functional assays.
Each ImmunoSpot® ELISPOT Kit includes all required buffers and washing solutions, and comes with our serum-free CTL-Test™ Medium, specially formulated for low background and high antigen-specific signal in T cell ImmunoSpot® assays. We also offer serum-free media for freezing and thawing cryopreserved PBMC.
To support your assay development, CTL provides peripheral blood mononuclear cells (PBMC) with well-defined characteristics. These serve as reliable positive controls and reference samples and include PBMCs with Th1, Th2, Th17 cytokine signatures, or antibody-secreting cell (ASC) reactivity against specific antigens (please inquire to learn more).
